Production of transgenic mice by pronuclear injection of DNA constructs
General information about strain nomenclature:
B6D2F1/J is a cross between C57BL/6J (B6) female and DBA/2J (D2) male. B6D2F1/J mice are heterozygous for B6 and D2 alleles at all loci in their genome. The CFT frequently uses this strain for transgenic/knockout mouse generation, as well as for other mutations.
Standard DNA microinjection service:
We inject DNA supplied by the investigator or generated by us into fertilized eggs. Our service covers the injection into fertilized eggs of max. 100 donor female mice. Instructions on preparing the DNA are available. The final purification step will be done by the CFT for plasmid-based transgenes.
Transgenic pups identified by the client will be delivered to the client after weaning (6 to 8 weeks after birth).
We require investigators to submit a restriction digest of at least 50 micrograms of the plasmid. The fragment should be purified like described in Pronuclear injection for the production of transgenic mice.
Please note: The transgenic status only refers to DNA integration. The CFT does not guarantee the expression of RNA or protein from the integrated DNA. The CFT will not furnish a guarantee of success.
