Our Genome Editing and Transgenesis Core Facility offers you a full-service for the generation of new genetically modified mouse lines at the SPF mouse facility of LIMES-GRC. In addition, we perform all transgenic standard services like rederivation of mouse lines, IVF, cryopreservation or microinjection into oocytes and blastocysts. We will also support you in genome editing projects in other species like Drosophila, Zebrafish or Xenopus.

Tell us your gene of interest and the desired modification and we will kick off your project. It will start with a short meeting to discuss your aims in more detail, and then we will suggest the best method and a project strategy. Within this outline, we will explain the advantages and disadvantages of CRISPR-mediated genome editing versus other strategies like gene targeting in ES cells and suggest what would be suited best for your knockout, conditional knockout or knock-in project.

As soon as the required, short animal license has been approved, we will start with the generation of your mice. During the project, you will get regular updates. At the end of the project, you will get a detailed report and confirmed, heterozygous mice (F1) which you can immediately use to start breeding for your specific scientific purpose. The generation of founder mice for simple knockout or knock-in projects via CRISPR takes about 4-6 months plus about two months for backcrossing to Bl/6. More complex knock-ins via ES cell targeting can take between 10-12 months.
